Prerequisite endocardial-mesenchymal transition for cardiac trabecular angiogenesis

We used single cell RNA sequencing (scRNAseq) to show that ventricular endocardial cells generate trabecular vessels through an angioEMT mechanism. We did E12.5, E13.5, E14.5 and E15.5 whole heart scRNAseq and intergated these 4 stage data together. We analyzed endothelial cell clusters and compared different subclusters in a stage-dependent diffence within subclusters, to find the potential mechanism of how ventricular endocardial cells generate trabecular vessels.

本研究借助单细胞RNA测序(single cell RNA sequencing, scRNAseq),证实心室心内膜细胞可通过血管内皮间质转化(angioEMT)机制生成小梁血管。我们对胚胎发育第12.5天(E12.5)、13.5天(E13.5)、14.5天(E14.5)及15.5天(E15.5)的全心脏样本开展单细胞RNA测序,并整合上述四个发育阶段的测序数据。随后我们针对内皮细胞簇展开分析,比较各亚簇间的发育阶段依赖性差异,以阐明心室心内膜细胞生成小梁血管的潜在分子机制。
