Homo Sapiens homology model of mutant of human cardiac troponin I. Homo Sapiens

Three heterozygous arginine to glutamine substitution at positions 98, 141 and 162 in TNNI3 protein were found in patient of hypertrophic cardiomyopathy. The crystal structure of TNNI3 was fragmented and co-ordinates of linking region from residues 137 to 146 were missing that also hosts one of the reported mutation. Hence, the missing residues 137 to 146 were generated with the help of available structure of TNNI3 using loop modeling program of MODELER 8.2 in the modeling environment of Discovery Studio 2.0. The generated model was used to build the model structure of three mutant TNNI3. The developed models were used to study the effect on interactions with other two subunits (troponinC and troponin T) of troponin complex.
