Single-nucleus RNA sequencing was used to profile transcriptional differences in mice in uninjured or neuropathic chronic pain conditions and in response to multiple sensory stimuli to identify cell subtype-specific changes in the transcriptome due to pain Overall design: We performed the 10x Genomics 3' gene expression assay (v3.1). Nuclei were isolated from the right amygdala of adult male mice that were either uninjured or 3 weeks following a left side spared nerve injury to induce chronic neuropathic pain. Uninjured mice were split into groups of 3 biological replicates that received either no stimulus or a light touch to the left hindpaw or a noxious hot water droplet to the left hindpaw. Mice with a spared nerve injury were split into two groups of 3 biological replicates that received either no stimulus or a light touch to the left (injured) hindpaw to induce chronic neuropathic pain. Nuclei capture and library preparation was perormed using 10x genomics.
