To ascertain whether the activation of AhR in intestinal epithelial cells (IECs) directly contributes to the enhanced production of Muc2, we generated conditional AhR knockout mice with deficiency in IECs (AhRΔIEC). Subsequently, the expression of genes involved in mucin production, along with the markers indicative of mature goblet cells and proliferation in the context of colitis, with or without I3C treatment was evaluated. We used microarrays to detail the global programme of gene expression to identify the distinct classes of up-regulated and downlregulated genes among the experimental groups. Mouse colon tissues from various experimental groups were collected for RNA extraction and subsequent hybridization onto Affymetrix microarrays.To investigate the role of AhR expression in intestinal epithelial cells (IECs), we employed both IEC-specific AhR knockout (AhRΔIEC) mice and wildtype (WT) mice.All mice were subjected to treatment with DSS and I3C, after which RNA was extracted from the following four groups: WT DSS, WT DSS+I3C, AhRΔIEC DSS, and AhRΔIEC DSS+I3C.
