EBV tegument protein, BNRF1, was found to inhibit a common host restriction factor, the SMC5/6 complex, during primary infection and viral lytic replication. RNAseq was used to identify host and viral transcriptome changes in BNRF1 knock-out P3HR1-Z/R-HTI cells, to get insights into other viral processes that is inhibited by the SMC5/6 complex. BNRF1-KO P3HR1-Z/R-HT cells were subjected to lytic induction by 4-HT and sodium butyrate, which were further used for RNAseq analysis. The P3HR1Z/R-HT cells expressing control sgRNA was used as the control.
