Objective To explore the regulation of TGF by extracts of Hedyotis diffusa- β/ The mechanism by which the Smad signaling pathway affects the proliferation, invasion, and Th1/Th2 cytokine expression of renal cancer cells in mice.Method Using mouse renal cancer Renca cells as the research object, after intervention with extracts of Hedyotis diffusa, the impact on the migration ability of mouse renal cancer cells was detected at the cellular level using scratch assay; Observing the effect of CCK-8 experiment on the proliferation ability of mouse renal cancer cells; A mouse kidney cancer model was established by subcutaneous inoculation under the right armpit, and the effect of extract of Hedyotis diffusa on the proliferation and invasion of renal cell carcinoma in mice was studied; Western blot analysis of TGF in mouse transplanted tumor tissue- β/ Smad pathway protein TGF- β、 Smad2 and p-Smad2 were detected; Detect the levels of IL-2 and IL-10 in mouse serum through the Elisa experiment to investigate whether the extract of Hedyotis diffusa has an inhibitory effect on Th1/Th2 renal cell carcinoma immune shift.Result The CCK-8 experiment showed that the extract of Hedyotis diffusa inhibited the proliferation ability of mouse renal cancer Renca cells (P<0.05); The scratch test results showed that the extract of Hedyotis diffusa effectively inhibited the healing ability of mouse renal cancer Renca cells in the scratch test (P<0.05); In vivo experiments have shown that by constructing a mouse kidney cancer model, it was found that the extract of Hedyotis diffusa can inhibit the growth of transplanted tumor volume in mice (P<0.05); HE staining test results: Compared with the model group, the extract group of Hedyotis diffusa and the pathway inhibitor group can inhibit the proliferation and migration ability of renal cell carcinoma in mice; Through Western blot experiments, it was found that compared with the model group, the extract group of Hedyotis diffusa and the pathway inhibitor group of TGF- β、 The expression of p-Smad2 decreased (P<0.05); Elisa's results showed that compared to the model group, the extract group of Hedyotis diffusa and the pathway inhibitor group showed an increase in IL-2 expression and a decrease in IL-10 expression (P<0.05).Conclusion The extract of Hedyotis diffusa may regulate TGF through regulation- β/ The Smad signaling pathway inhibits the drift of Th1 to Th2 in mouse renal cancer, thereby inhibiting the proliferation and migration of mouse renal cancer cells both in vitro and in vivo.
