We employed whole genome microarray expression profiling as a discovery platform to identify gene expression changes after DHTKD1 knock-out RNAs of spinal cords of 8-weeks-old wild-type (n=3) and DHTKD1 knock-out mice (n=3) were used to identify gene expression. Spinal cords were dissected and frozen into liquid nitrogen immediately. Total RNA was extracted and checked for a RIN number to inspect RNA integration. Qualified total RNA was further purified, amplified and labeled. Labeled cRNA were further purified. Each slide was hybridized with 1.65μg Cy3-labeled cRNA for 17 hours at 65°C in rolling hybridization oven. Then, slides were washed and scanned by Agilent Microarray Scanner with default settings.
