TGFB in bone marrow dysfunction in diabetes

We performed a microarray analysis to understand the mechanisms of TGFB1-mediated inhibition of adipogenesis on cultured human primary bone marrow-derived mesenchymal progenitor cells (bm-MPCs). Our goal was to identify targetable pathways that mediate the effect of TGFB1. Human bone marrow-derived mesenchymal progenitor cells were cultured at a density of 40,000 cells/cm^2. We cultured bm-MPCs in normal growth media (Control), adipogenic differentiation media (ADP), adipogenic differentiation media with TGFB1 (10 ng/mL), and adipogenic differentiation media with TGFB1 + TAK1 inhibitor (10 umol/L). Total RNA was purified and transcriptome-wide analysis of gene expression was performed by Clariom S Assay.
