Gene expression profile of individual CFU-GM colonies identified as uniallelic TET2 mutant versus TET2 wild type was analyzed via RNA-Seq to verify the impact of TET2 editing on gene expression in the rhesus macaque clonal hematopoiesis model. Overall design: Mobilized CD34+ cells were treated with CRISPR/Cas9, followed by transplanted autologously to generate a non-human primate model of clonal hematopoiesis. CD34+ HSPCs were enriched from the bone marrow at 15 months post-transplantation and plated for CFUs. At 14 days, individual CFUs were picked and genotyped, and three TET2 wild type and three TET2 uniallelic CFU-GM CFUs with predicted loss-of-function mutations were analyzed via RNA-Seq.
