A genotyping-by-sequencing (GBS) experiment with 96 trees of several coastal and interior Douglas-fir (Pseudotsuga menziesii) provenances was performed. DNA of the trees was digested with one restriction enzyme (ApeKI, single digestion). Fragmented and barcoded DNA were pooled and then sequenced on one lane of a HiSeq1000.
