Single-Cell Profiling and Cell-Cell Communication Analysis of Splenic CD11c? Cells in Wild-Type Mice

Single-Cell RNA-sequencing and Cell-Cell Communication analysis of Splenic CD11c? Cells from Wild-Type Mice Overall design: We conducted single-cell analysis of splenic CD11c? cells from wild-type mice to characterize immune cell heterogeneity and intercellular communication. CD11c? cells were enriched using MACS beads and labeled with a multiplexing kit before capture on the BD Rhapsody Single-Cell Analysis System. Mouse macrophages (MMMs) and Langerin? cDC1s were identified and subsetted using Seurat, and cell-cell communication networks were inferred using the CellChat R package. This approach enables detailed mapping of immune interactions within the splenic microenvironment.
