The 3D organization of the genome has recently been considered to have essential impact on nuclear functions such as gene expression and epigenetic modifications. In this project, we develop a novel application of next-generation sequencing technology to analyze the 3D genome organization in the cell nucleus, in combination with a transposon-based genome engineering approach. Most DNA transposons have a tendency to jump around locally in the genome, referred to as âlocal hoppingâ tendency. By using next-generation sequencing technology, we globally identify the transposon insertion sites in a genome-wide manner, and investigate the correlation between the distribution of transposons and the spatial organization of the genome.
