The presented study aimed at the development of a protocol for efficient culture of rat neonatal glial cells (microglia, astrocytes, and oligodendrocytes) in oxygen concentrations relevant to the nervous tissue (5%). The cells are cultured in media without supplement addition. The application of active biomolecules for coating culturing surfaces might be useful for mirroring physiological cell interactions with extracellular matrix components. Applying additional procedures, like transient oxygen and glucose deprivation (OGD), allows to mimic in vitro the selected pathophysiological conditions e.g. perinatal asphyxia.In the study we evaluated proliferating rate of neonatal oligodendrocytes in vitro in control (ctr) and OGD-subjected (OGD) cultures. Results of the analyses of the data are presented in Figure 5.G.We also analysed the number of neonatal rat astrocytes cultured on surfaces covered with different biomolecules like laminin (LAM), fibronectin (FBR), Extracellular Matrix components (ECM Gel) and poly-L-lysine (PLL) in control (ctr) and OGD conditions. Results of the analyses of the data are presented in Figure 6.D.We measured EAAT1 expression (integrated density) by astrocytes attached to various biomolecules using ImageJ software. Results of the analyses of the data are presented in Figure 7.J.
