The V2a neurons contribute significantly to recovery of locomotor and respiratory function after injury in animal models. We used single nucleus RNA sequencing of enriched cervical V2a neurons to analyze the diversity of these neurons and their response to spinal cord injury. Overall design: Nuclei from Chx10Cre/+; ROSA26CAG-LSL-Sun1-sfGFP-Myc/+ mouse cervical spinal cords were isolated from uninjured mice or 5 days post C2 hemisection spinal cord injury. Fluorescence-activated nuclei sorting (FANS) was used to isolate V2a (GFP+) nuclei. The sorted nuclei were then subjected to single nucleus RNA sequencing using 10X Chromium 3v3.
