This dataset investigates the role of Polyglutamine-binding protein 1 (PQBP1) in protecting testicular germ cells from heat stress. Our hypothesis is that PQBP1 is crucial for forming protective stress granules (SGs). The data was collected from wild-type and PQBP1-Y65C mutant mice before and after testicular heat shock. It includes RNA-Seq transcriptome data, immunofluorescence images of SG formation, sperm motility analysis (CASA), and histological data. The findings show that the PQBP1 mutation impairs SG assembly, dysregulates the transcriptional response to heat, and significantly reduces sperm motility after heat stress. This data indicates that PQBP1 protects male fertility by orchestrating an effective cellular stress response and can be used to explore mechanisms of heat-induced infertility.
