H3K27ac ChIP-seq analysis was performed using zebrafish normal thymus and primary tumors to analyze potential regulatory elements activated by IRF4 overexpression. Overall design: A transgenic zebrafish line expressing human IRF4 and mCherry genes under zebrafish lck promoter and a control line expressing mCherry gene only under zebrafish lck promoter were established. Normal thymus from control line and tumor cells from IRF4-transgenic line were sorted and crosslinked with formaldehyde for 10 min. DNA was enriched by chromatin-immunoprecipitation (ChIP) and analyzed by BGISEQ-500. A sample of whole cell lysate (WCL) was sequenced and used as the background to determine enrichment. ChIP was performed using an antibody against H3K27ac antibody (Abcam, ab4729).
