To clarify the global gene expression alteration in primary CD34+ AML cells after BCAA metabolism inhibition, CD34+ AML cells were treated with or without gabapentin in vitro. After 9-12 hours of culture with or without gabapentin, FACS-purified CD34+ AML cells were subjected to transcriptome analysis. To clarify how BCAA metabolism affect the gene expression of human CD34+ AML cells, CD34+ AML cells from 4 independent patients were treated with or without 40mM gabapentin, a specific BCAT1 inhibitor for 9-12 hours. After BCAA metabolism inhibition by gabapentin, CD34+ AML cells were purified and total RNA was extracted from control and treated samples.
