Whole genome sequencing of E. coli BW25113 strains with edited topA gene

Topoisomerase I of Escherichia coli (EcTopoI) is encoded by the topA gene. EcTopoI relaxes negatively supercoiled DNA and is thought to maintain the steady-state level of supercoiling by compensating the activity of another topoisomerase - DNA gyrase. By recombineering, we constructed E. coli strains with topA_delta11, topA_delta14, and topA_delta30 mutations that led to the production of EcTopoI lacking, correspondingly, an 11 kDa portion of CTD, the entire 14 kDa CTD, or a longer 30 kDa fragment that includes both the CTD and the Zn-binding domain. To investigate if these deletions led to acquiring of compensatory mutations we performed whole-genome sequencing of the strains.
