The single-cell RNA sequencing of Panax species, yielding commercially valuable ginseng products, is of great significance to their molecular breeding. But this study has not yet been reported. Herein, scRNA-seq of three Panax species radicles, specifically P. notoginseng, P. ginseng, and P. quinquefolium, was performed. By integrated analysis, we constructed a single-cell transcriptome atlas of Panax radicles and then reconstructed developmental trajectory of endodermal cells and elucidated transcriptional regulation networks affecting endodermis development, ginsenoside biosynthesis, and cold tolerance.
