RNA sequencing of primary mouse ileal organoids treated with bile acid pools with various compositions

Bile acids act as ligands for several nuclear receptors expressed in the small intestine, especially the terminal ileum. Bile acid pool composition can be influenced by health and disease states. Here, we used RNA-seq to investigate the effects of altering bile acid pool composition on gene expression in ileal organoids. Primary mouse ileal organoids were treated with vehicle or a designer bile acid pool for 24h. 4 distinct bile acid pools were used, which varied based on the species we modeled (human or mouse) and the proportion of 12a-hydroxylated bile acids (10% or 90%). 3 samples from each treatment group were submitted for RNA sequencing.
