RNA-seq of Ficus carica cultivars Sabrutsiya Rozovaya, Aromatnyy Nikitskiy, Nairanneyshiy Fioletovyy

RNA-seq analysis was conducted using RNA samples isolated from leaves of Ficus carica cultivars Sabrutsiya Rozovaya, Aromatnyy Nikitskiy, Nairanneyshiy Fioletovyy from Nikita Botanical Gardens fig trees collection. We used plants grown under three alternative conditions - in vitro culture, greenhouse plants (in vitro), and open field trees (ex-situ) to retrieve changes in genes expression pattern during plant adaptation from in vitro culture to ex-situ state. Total RNA was isolated using the standard Nucleospin RNA Plant protocol (Macherey-Nagel, Germany). Sequencing was conducted on Illumina NextSeq 500 by NextSeq 500 Mid Output v2 Kit (150 cycles) according to standard protocols with 150 bp in the forward and reverse directions.
