RNA sequencing of leaf tissue from Portulaca amilis and P. oleracea over the course of a water limitation experiment that induced Crassulacean Acid Metabolism (CAM) in both species. P. amilis were germinated in a growth chamber with the following environmental conditions: 00:00-06:00, dark, 22 degrees C; 06:00-20:00, lights on, 25 degrees C; 20:00-23:59, dark, 22 degrees C. Six biological replicates per species were regularly bottom watered and the mean photosynthetically active radiation (PAR) at plant level was 385 mol * m-2 * s-1 during the 14 hour photoperiod. After plants had reached maturity and branched sufficiently (3-4 weeks), water was withheld and significant increases in nocturnal titratable acidity were observed in P. amilis and P. oleracea after 8 and 9 days, respectively. Leaf tissue was collected from all six biological replicates for titratable acidity assays and bulk RNA at 10:00, 16:00, 22:00, and 04:00 (the following day) on day 0 and days 8 and 9 of water withholding.There are 92 samples in total:- 6 P. amilis replicates x 2 sampling periods x 4 sampling times = 48 samples- 6 P. oleracea replicates x 2 sampling periods x 4 sampling times - 4 library failures = 44 samples
