Bulk RNA-seq: Secondhand Smoke Exposure on 4 and 6 days Before Delivery on mice

Placental tissue was collected from SHS-exposed and control mice at necropsy for transcriptomic profiling. Total RNA was extracted using the Direct-zol RNA MiniPrep Plus Kit with TRI Reagent, including on-column DNase I treatment to eliminate genomic DNA. mRNA was enriched with poly-dT magnetic beads, converted to cDNA, and processed through end polishing, A-tailing, and Illumina adapter ligation. Libraries were size-selected, amplified, and quality-checked by Qubit fluorometry, qPCR, and Bioanalyzer analysis before paired-end sequencing on an Illumina NovaSeq platform.
