The inPOSE method integrates transgenes into bacterial genomes by homologous recombination, targeting insertion sequence (IS) elements residing in the genome. As a second step, the transposition of the modified IS elements, carrying the cargo gene is induced. The resulting copy/paste type if transposition leads to the copy-number amplification of the integrated transgenes. This process is monitored in four samples: B0 is the starting E. coli strain, B1, B2 and B3 are the strains that have undergone 1, 2 or 3 rounds of transposase induction, respectively.
