Using a custom capture panel designed for the 610 human tRNAs (as annotated in GtRNAdb), DNA containing tRNA-encoding genes was isolated from 84 individuals and sequenced.. tRNA variation in human samples

The Nextera Custom Enrichment kit (Illumina) was used to capture genomic regions corresponding to the 610 tRNA-encoding genes. Coordinates were obtained from GtRNAdb 2.0 and chromosome scaffold coordinates were obtained from the UCSC genome browser using the February 2009 GRCh36/hg19 genome build. Sequences were submitted to the Illumina Online Design Studio for target-probe design. Eighty-four individuals were sequenced in this study. Forty-eight individuals with hypertriglyceridemia (HTG) had plasma triglyceride levels greater than 10 mmol/L with no genetic explanation. The other 36 individuals were controls with triglyceride levels less than 2 mmol/L. DNA was isolated from 4 mL of blood using the Puregene DNA Blood Kit (Gentra System, Qiagen Inc.). Libraries containing 12 samples each were prepared as described in the Nextera Rapid Capture Enrichment protocol and sequenced on an Illumina MiSeq.
