Landoltia punctata cDNAs were reversed transcribed from polyA-selected mRNAs. SfiIA and SfiIB linkers were added to the cDNAs via the primers used to synthesize first and second strand cDNAs. The cDNAs were cut with SfiI to generate SfiI sticky ends, and the cDNAs were directionally ligated into SfiIA and SfiIB-cut plasmid vector pTriplEX2. Recombinant plasmids were transformed into E. coli strain DH5a
