This graph shows that optimized doses of phosphonoformic acid (PFA) from 0.25-0.75 mM for either a one-hour pre-treatment (PRE) prior or continual exposure during 72 hours of incubation in the presence of zoledronate do not rescue the 50 microM zoledronate (ZOL)-treated RAW264.7, RANKL-differentiated osteoclasts. "MEDIA" control percent viability was mildly but significantly lower than the viability of the combined ZOL + PFA vehicles control (blue bar); consequently, the vehicles had a positive effect on viability. All other percent viabilities for the other conditions were expressed relative to the ZOL + PFA VEHS' viability. Error bars are SEM and the asterisk indicates ZOL-treated RAW264.7, RANKL-differentiated osteoclasts are significantly less viable than ZOL + PFA VEHS control cells. N=6.
