To investigate site-specific methylation for RNA methylation by a methyltransferase and its ancestral proteins, we performed Oxford Nanopore direct RNA sequencing (DRS) to characterize the site specificity of RNA methylation in three different methyltransferases. The RNA substrate (1.8 kilobases, 1468 adenine-containing 5-mer motifs) used for methylation assays was methylated using each enzyme and sequenced using an Oxford Nanopore MinION device.
