To select an inhibitory peptide targeting BORIS, the N-terminus of BORIS between amino acids 1 and 258 (BORIS-N1-258) was used as an antigen for the selection from the Ph.D.-12 phage display peptide library. The peptide with the highest affinity was fused with the HIV-1 TAT peptide to penetrate the cell membrane and was designated BORIS-targeted peptide (BTApep-TAT). Validation of the effect of BTApep-TAT on non-small-cell lung cancer (NSCLC) progression indicated that this inhibitor targeted BORIS. To investigate the targeting specificity of BTApep-TAT toward BORIS, the transcriptomes of H1299 cells treated with BTApep-TAT and subjected to BORIS knockdown by two siRNAs were compared by high-throughput sequencing.
