This study contains 93 samples in two subsets: 33 Black_yellow_astatos (look for parallel evolution of black males from yellow ancestors in multiple crater lakes), and 60 samples from Lake Rukwa (Blue/yellow ecotypes plus riverine flowing in). These projects concern data from experiments undertaken as part of the Durbin group’s long-term research into the evolutionary genomics of the Lake Malawi cichlid fish radiation. All samples will have DNA extracted at WSI from ethanol preserved fin clips, for which there is a standard 96-well QiaSymphony solution. Libraries will be standard Illumina WGS or Haplotagging, and sequencing will be on Illumina Novaseq S4 PE150. We wish to sequence to approximately 15x coverage. The Malawi radiation has created several hundred species of cichlid fish that have low genetic divergence (0.15-0.25%) but high phenotypic divergence. Results so far from us and others have identified multiple loci under selection and/or genetically associated with adaptive traits. Loci found in wild outbred studies can be further studied in crosses and vice versa. The proposed sequencing will follow up previous results and search for new loci.
