In order to investigate the effect of in vitro hyperglycemia on gene expression of fpEC, cells were treated with hypergylcemia (25mM) for 24h. 5 different isolations of primary fpEC (800,000) were split in two gelatin coated 75cm2 culture flasks in EBM medium. After 24h, medium was replaced either by normoglycemic EBM (5.5mM D-glucose) or with EBM containing 25mM D-glucose. After 24h, RNA was isolated using TRI REAGENT. 5µg RNA of all control conditions and 5µg of each hyperglycemic treatment was pooled and the pools used for the microarray experiment.
