To evaluate the genetic effects of low dose radiation in the natural environment on germ line cells, de novo mutations in the next generation were surveyed for Japanese cedar and flowering cherry trees inhabiting radioactive contaminated area by the Fukushima Dai-ich Nuclear Power Plant accident. Direct comparison of nucleotide sequences was made between parent and next generations of haploid megagametophytes and zygote seeds/seedlings for Japanese cedar and cherry trees, respectively. The double digest RADseq libraries were constructed using restriction enzymes (PstI and SphI for Japanese cedar and MseI and PstI for flowering cherry), and the libraries were sequenced using HiSeq X with 2x151-bp paired-end reads by Macrogen Japan Corp.
