Effects of substrates on gene expression of human endometrial organoids

To investigate the effects of different substrates on human endometrial organoids , we compared the transcriptomes of EMO cultured within Matrigel and collagen-based gels. Overall design: We cultured human endometrial organoids in Matrigel or collagen-based gels (70% type-I collagen and 30% matrigel) for 7 days. Human endometrial organoids were established and maintained in the ECSY medium. The composition of the ECSY medium was as follows: DMEM/F-12 (Life Technologies) supplemented with 1% ITS-X supplement, 0.15% BSA (FUJIFILM Wako), 1% Knockout Serum Replacement (KSR, Thermo Fisher Scientific), 200 mM L-ascorbic acid (FUJIFILM Wako), 50 ng/ml EGF, 2 µM CHIR99021, 2 µM SB202190, and 10 µM Y27632. We used 2 independent organoids from 2 donors.
