We sequenced small RNAs from shoot and root of 13 days old seedlings of heat tolerant (Nagina 22) and susceptible (Vandana) cultivars of Oryza sativa to analyze heat responsive microRNAs. Seedlings were treated with control temperature (30°C/24°C, day/night) and high temperature (42°C/36°C, day/night) for short (24h) and long (5 days) duration. To get the recovery samples, high temperature treated seedlings were shifted back to control temperature for 24 h. Sixteen small RNA libraries were constructed from control shoot, control root, short duration stress (SDS) shoot, SDS root, long duration stress (LDS) shoot, LDS root, recovery shoot, and recovery root tissues of both the cultivars.
