Domestic cats and dogs are common household pets, with cancer a common cause of morbidity and mortality in these companion animals. Since the feline oncogenome remains uncharacterised, we sequenced 493 feline tumor-normal tissue pairs comprising 13 tumor types, focusing on the feline orthologs of ~1,000 human cancer genes. <i>TP53</i> was the most frequently mutated gene identified across all tumor types, and one of 31 driver genes identified. Together with identifying somatic copy number alterations, mutational signatures, viral sequences, and tumor-predisposing germline variants, our study provides a comprehensive picture of the feline oncogenome. Importantly, this study demonstrates key similarities with the human oncogenome, confirming the cat as a valuable model for comparative studies, and identifies potentially actionable mutations, which align with a ‘One Medicine’ approach. The files in this dataset are the Supplementary Tables to accompany the manuscript for this study.
