The Amish and Hutterites are U.S. farming populations with remarkably similar lifestyles. However, the Amish follow traditional farming practices, while the Hutterites employ modern farming techniques, and also show striking differences in asthma prevalence. Little is known about immune responses underlying these differences. To address this, we measured environmental exposures, genetic ancestry, and immune profiles in 60 Amish and Hutterite schoolchildren. We further collected indoor house dust to measure allergen, endotoxin, and microbiome composition, and immune and airway responses to house dust extract were tested in murine models of experimental allergic asthma.
