The orange-spotted grouper (Epinephelus coioides) is a protogynous hermaphroditic fish, and its mechanism of sex reversal is still unknown. In this study, the orange-spotted grouper was induced to change sex from female to male by 17alpha-methyltestosterone implantation. During the process of sex reversal, two mainly oocyte types (primary-growth stage oocytes, cortical-alveolus stage oocyte) were isolated by laser capture microdissection. Subsequently, transcriptomic analysis for the isolated cells was conducted. A series of genes were used to verify and investigate the expression patterns in sex reversal. Furthermore, we also analyzed the expression of the set of genes involved with the pathways of signal regulating pluripotency of stem cell and steroid metabolism in the orange-spotted grouper.
