To further development of our gene expression approach, we have employed whole genome microarray expression profiling to identify genes related to VEGF in MDA-MB-231 breast cancer cell line. Overall design: Total RNA of MDA-MB-231, VEGF-knocked out MDA-MB-231, recombinant VEGF(rhVEGF) treated VEGF-knocked out MDA-MB-231, soluble NRP1 overexpressed MDA-MB-231 cells were used. RNA was quantified using a NanoDrop-1000 spectrophotometer and quality was monitored with the Agilent 2100 Bioanalyzer (Agileng Technologies, Santa Clara, CA). VEGF was knocked out using CRISPR/Cas9 system. Soluble NRP1 works as VEGF-TRAP. Cell were cultured for 48 hours before RNA extraction. rhVEGF treatment was done for 48 hours.
