We report the development of precision guided sterile insect technique (pgSIT) in Aedes aegypti. We engineer strains that express either Cas9, or a set of gRNAs guiding simultaneous disruption of genes essential for either male fertility or female flight. When these strains are genetically crossed, resulting progeny are sterile males and flightless females, which can compete, suppress, and eliminate mosquito populations.
