Disrupting tRNA modifications to target mitochondrial vulnerabilities in drug-resistant leukemia cells

To comprehensively identify cancer-relevant RNA modifications and their regulators, we screened all 150 known human RNA modifying proteins across 18 different normal and cancer lines using a CRISPR-based genetic knockout system. Gene expression profiling of 2 primary human healthy lines (normal human epidermal keratinocytes and normal human epidermal melanocytes) and 16 different cancer cell lines including acute myeloid leukemia, head and neck squamous cell carcinoma, non-small-cell lung carcinoma, melanoma, and multiple myeloma.
