Multiple myeloma (MM) cell lines were transduced with Doxycyclin-inducible retroviral vectors encoding two separate shRNAs targeting the NF-kappaB transcription factor RELA (shRELA) and Renilla (shRenilla) as control. At day 3 post Doxycycline induction cells with shRNA knockdown (identified as YFP, DsRED double positive cells) were isolated by fluorescent activated cell sorting. RNA was isolated and reverse transcribed for RNA-sequencing analysis. For each shRNA, triplicate cultures were processed (n1-n3). All MM cell lines require RELA for cell growth or survival. The data are useful for the determination of the oncogenic biological programs controlled by the RELA transcription factor in MM with oncogenic NF-kappaB activation, which is expected to have implications for the development of targeted therapies against subtypes of MM.
