Festuca pratensis strain:HF27, B1416 Transcriptome or Gene expression

Non-cold treated plants were grown at 20/12*C day/night, 16/8h light/darkness with light intensity 240 µmolm-2s-1 for two weeks. Cold treated plants were grown at 20/12*C day/night, 16/8h light/darkness with light intensity 240 µmolm-2s-1 for two weeks and then subjected to cold acclimation at 1*C. In all experiments tissues were sampled from three clonal plants and pooled in equal ammounts. Total RNA (100mg) was isolated from crown, stem and leaf tissues using the RNeasy Mini kit (Qiagen, Hilden, Germany). Finally, RNA from all tissues were pooled. cDNA was synthesized using MINT-Universal cDNA synthesis kit (Evrogen, protocol-II) and sequenced using a single-end sequencing protocol on Roche 454 Titanium.
