The goal of this study is to determine how the loss of the transcription factor Sox9 affects the molecular profiles of adult astrocytes from the olfactory. We performed RNA-sequencing on wildtype and Sox9 knockout (KO) astrocytes from the olfactory bulb and analyzed the molecular signatures of Sox9 KO astrocytes compared to the wildtype control. AAV-GFAP-iCre-P2a-TurboRFP was injected into the olfactory bulbs of Sox9fl/fl mice or C57BL/6J mice to generate astrocyte-specific Sox9 knockout and control mice. FAC sorted astrocytes from the olfactory bulbs were collected in triplicate from our viral Sox9 KO mouse line and control mice 4 weeks after the viral injection. Sox9 KO and control astrocytes were molecularly profiled using RNA-sequencing.
