Two healthy donors PBMCs were stained with fluorescent antibodies for cell sorting of MAIT cells (CD3+, CD161+, MR1-5-OP-RU tetramer+), NK cells (CD3-, CD19-, CD14-, CD16+ CD56dim), gamma-delta T cells (CD3+, TCRgd+) and iNKT cells (CD161+ iNKT+) using FACS. Each donor was analyzed ex-vivo, after 24 hours of incubation with Mtb lysate, or after 24hours of incubation in plain serum.
