Construction of Engineered Bacteria Secreting Bacteriocin Lmo2776 and Validation of its Effect in Mice

We integrated the bacteriocin Lmo2776 module into the genome of E. coli Nissle 1917 (EcN). Yielding the Lmo2776-secreting strain EcN-2776. we selected high-fat mice as the model for in vivo validation to compare the differences in intestinal microbiota after gavage, At the same time, a control group of wild-type mice was established. We collected intestinal contents for 16s sequencing analysis, 16S rRNA gene amplicon sequencing indicated that intestinal secretion of Lmo2776 adjusted the gut microbial composition of HFD-fed mice. The results showed that the constructed engineered strain EcN-2776 produced an effect and exerted corresponding functions in mice.
