To compare The pattern of Gene expression by 7-hydroxymethotrexate-resistant human leukemia cells reveals changes designed to preserve intracellular levels of folate and nucleotide biosynthesis We used microarrays to detail the global programme of gene expression underlying cellularisation and identified distinct classes of up-regulated genes during this process. Keywords: Resistant and non-resistant MOLT4 cell lines Treatment of human T-cell leukemia lines to the major plasma metabolite of methotrexate (MTX), 7-hydroxymethotrexate (7-OHMTX), resulted in an acquired resistance as a results of complete loss of folypolyglutamate synthetase (FPGS) activity. To identify the genes associated with MTX and 7-OHMTX resistance, we analyzed the patterns of gene expression profiles by leukemia sublines and compared with parent cells.
