Sequencing of Tn5 transposon insertion library of E. coli K-12 W3110 subjected to FRET-To-Sort

We combined a FRET-based c-di-GMP biosensor toolbox with a barcoded Tn5 transposon library and cytometry-based cell sorting to develop FRET-To-Sort, a new application for systematic characterization of gene networks regulating levels of FRET-detected small molecules. Applied to planktonic E. coli cells, FRET-To-Sort allowed a genome-wide mapping of c-di-GMP regulatory networks.
