The experimental aim was to isolate differences in gene expression in response to innate immune stimuli between WT and LRRK2 KO mouse macrophages. Cells were treated with 100 ng/ml of LPS or 10 ug/ml MDP for two hours, after which mRNA was extracted and RNA sequencing performed. 3 replicates of the activation experiments were conducted on macrophages derived from three wild-type and LRRK2 knockout mice that were divided and treated with either control medium, LPS containing medium, or MDP containing medium.
