We investigated gene expression changes of glucose deprived MCF-7 and T47D breast cancer cells supplemented with either 10 mM or 25 mM BHB. Glucose deprivation revealed numerous differentially expressed genes indicating an involvement of the Hippo pathway in MCF-7 cells and the NRF2-Ferroptosis axis in T47D cells. Beta-hydroxybutyrate had limited impact on breast cancer cells and differentially expressed genes were not associated with any pathways following pathway enrichment analysis. Overall design: Two established human breast cancer cell lines were used for this analysis, MCF-7 and T47D. Cells were treated in either 225 mg/l glucose (5%) or 4.5 g/l glucose (100%). Cells treated with 225 mg/l glucose were additionally treated with no, 10 mM or 25 mM beta-hydroxybutyrate. Each condition was analyzed in duplicate except the comination glucose5%_BHB10mM in MCF-7 cells since one of the replicates failed library construction.
