RNA-sequencing of in vitro differentiated shRenilla (control) and shClns1a transduced pTh17 cells. CD4+ T cells isolated from the spleen of 2D2 mice were activated by plate-bound aCD3 (1 ug/ml) and aCD28 (1 ug/ml) in the presence of IL1b (20ng/ml), IL6 (20ng/ml) and IL23 (20ng/ml), aIL4 (5mg/ml). 24 hours post activation, cells were transduced with shRenilla (control) or shClns1a. 48 hours post activation, cell were detached and cultured in the presence of IL1b (20ng/ml), IL6 (20ng/ml), IL23(20ng/ml), and aIL4 (5mg/ml) for an additional 72 hours. Cells were restimulated with aCD3 (1 ug/ml) and aCD28 (1 ug/ml) for an additional 0, 6 or 24 hours. Transduced cells were enriched by FACS.
